<dc:description>
Introduction: Surgical trauma and the subsequent foreign body response can lead to connective tissue formation around cochlear implant electrodes, potentially impairing electrical stimulation. Methods: Model electrodes made from platinum-iridium wire were fabricated to approximate the surface area of conventional cochlear implant electrodes. The electrodes were embedded in silicone and coated with PVDF-TrFE fiber mats. Methods to promote wetting were investigated, and coatings produced using three different electrospinning durations (6, 12, and 18 minutes) and three different polymer concentrations were electrically characterized. In addition, fiber mats were loaded with dexamethasone at concentrations of 1%, 5%, or 10% (w/w) using either a blend solution or coaxial electrospinning, and drug release into artificial perilymph was monitored over two weeks. Results: Model electrodes were successfully fabricated, embedded, and electrically characterized prior to fiber mat coating. Wetting was promoted by supplementing 30% ethanol to the conductive solution. Electrical impedance increased with fiber mat application and with increasing electrospinning duration and polymer concentration. Dexamethasone release from the fiber mats was slow, with relative releases not exceeding 4% after two weeks for any of the concentrations or variants investigated. Greater dexamethasone release was observed from blend fiber mats than from the corresponding coaxial variants. Discussion: The increase in impedance caused by the additional fiber mat layer can be modulated by varying fiber mat morphology. Ethanol can promote wetting, although its use is limited to applications in which biocompatibility is not required. Although long-term release studies are needed to further evaluate drug release from the loaded fiber mats, the short-term results indicate their potential for sustained drug delivery. Overall, surface modification of cochlear implant electrodes with electrospun fiber mats may provide a strategy to mitigate the electrical insulation associated with post-implantation connective tissue formation.
</dc:description>
