<dc:description>
Hepatocellular carcinoma (HCC) is characterized by poor prognosis and limited therapeutic options, necessitating novel agents with selective efficacy and minimal toxicity. This study investigated the anticancer properties of saponin fraction (SPS) extracted from Holothuria leucospilota against HepG2 hepatocarcinoma cells. SPS were extracted and separated using chromatographic techniques. Cytotoxicity was assessed via MTT assay in HepG2 and normal THLE-2 hepatocytes to determine the IC50 and selectivity index (SI). Functional assays, including wound healing, transwell migration, colony formation, apoptosis (Annexin V/PI staining), and luciferase reporter assays, were conducted. Gene expression of Wnt/β-catenin pathway components was quantified using qPCR. SPS displayed potent cytotoxicity against HepG2 cells (IC50 = 1.14 μg/mL) with high selectivity (SI = 2.04). SPS significantly inhibited migration, invasion, and colony formation while inducing apoptosis in a dose-dependent manner. Mechanistically, SPS reduced Wnt/β-catenin signaling activity, as evidenced by decreased SuperTOP/FOP ratios and downregulation of β-catenin, Cyclin D1, c-Myc, Wnt3a, LEF1, and MMP7 transcripts. SPS exerts multifaceted antitumor effects in HCC, including induction of apoptosis, suppression of metastatic traits, and attenuation of Wnt/β-catenin signaling. Generally, SPS exhibited significant antiproliferative, anti-migratory, and pro-apoptotic effects in HepG2 cells, via modulation of the Wnt/β-catenin signaling pathway.
</dc:description>
