<dc:description>
This dataset accompanies the manuscript “Recovering elevational plant diversity patterns from Malaise trap bycatch using a dedicated filtering workflow”.

The study used ITS2 metabarcoding to investigate plant DNA recovered from Malaise trap preservative ethanol in the Gyirong Valley, central Himalayas. Sampling covered 13 sites along an approximately 2,400 m elevational gradient during six collection periods from 19 May to 17 July 2024. A total of 77 samples were analysed; sample E18 was lost during transport.

The dataset contains four components:

1. RAW-sequences: 154 compressed FASTQ files containing paired-end reads for 77 samples. R1 and R2 identify the forward and reverse reads, respectively. These files contain both target plant and non-target sequences.

2. rarefied sequences: QIIME 2 artifacts containing the rarefied OTU abundance table and the corresponding representative ITS2 sequences.

3. sequence annotation before LULU curation: An Excel workbook containing 1,386 manually annotated OTUs, their representative DNA sequences, taxonomic assignments and read counts across the 77 samples.

4. sequence annotation after LULU curation: An Excel workbook containing 487 vascular plant OTUs retained after LULU curation and subsequent manual review, with representative sequences, taxonomic assignments and sample-level read counts. This dataset contains 3,045,848 reads and precedes the five-step ecological filtering workflow described in the manuscript.

Each annotation workbook includes a Readme sheet, an OTU-by-sample abundance matrix and a sample-level table listing positive detections. The Readme explains sample identifiers, collection periods and measured sampling-site elevations. In sample identifiers such as A18, the letter indicates the collection period and the numeric suffix identifies the sampling station.

Abundance values are sequencing read counts, not counts of individual plants. DNA sequences in the Excel workbooks are OTU representative sequences, whereas the original sequencing reads and their quality scores are provided in the FASTQ files.
</dc:description>
