<dc:description>Amphibians can easily live in an abundant microbial environment on account of their specific organic secretions providing a unique source of bioactive peptides. Antimicrobial peptides (AMPs) are considered as feasible alternatives to conventional antibiotics due to their low resistance and broad spectra against bacteria, fungi and even cancers. <br>In present study, a novel natural dermaseptin peptide, preliminarily named as QUB-2437, was identified from the defensive skin secretion of the Jaguar Leaf Frog, Phyllomedusa palliata. The cDNA library was constructed from skin secretion-derived mRNA. The encoded amino acid sequence of mature peptide was deduced as GLWSTIKNVGKEAAIAAGKAVLGAL-NH2 by molecular cloning technique. Subsequently, QUB-2437 was synthesised using SPPS, and RP-HPLC was employed to purify the synthetic product. The molecular weight of QUB-2437 was confirmed by MALDI-TOF MS. Afterwards, antimicrobial assays, MTT cell proliferative assay as well as haemolytic assay were carried out to evaluate its bioactivity and cytotoxicity. The novel QUB-2437 exhibited different antimicrobial activities against three strains of microorganism including S. aureus, E. coli and C. albicans with MIC values of 16, 8 and 128 μM, and MBC values of 16, 8 and 256 μM, respectively. It was also discovered that there was an inhibition against four human cancer cell lines NCI-H157, U-251 MG, PC-3 and MCF-7. However, a relatively strong cytotoxicity on horse erythrocytes was observed as well. QUB-2437 possessed up to 5% haemolytic potency at concentration of 8 μM.<br>The above results indicated that structural modification would be required to reduce the cytotoxicity of the natural dermaseptin peptide for future clinical application. <br></dc:description>
