<dc:description>With the global abuse of antibiotics, many bacteria have developed resistance to multiple antibiotics, worsening the effectiveness of drug treatments. Consequently, there is a growing need to discover new antimicrobial agents. Frog skin antimicrobial peptides (AMPs) have shown promising potential as therapeutic drugs due to their natural effectiveness against a wide range of pathogens and their unique mode of action, offering a novel solution to the growing problem of antibiotic resistance. This thesis focuses on the peptide QUB-2607, which was found in the frog skin secretion of Phyllomedusa sauvagii. This study aims to identify a bioactive peptide from the skin secretion of the waxy monkey leaf frog, native to South America, Phyllomedusa sauvagii, and to evaluate its biological activity. Its sequence was obtained through molecular cloning. The peptide was synthesised by solid-phase peptide synthesiser, purified by reverse-phase high-performance liquid chromatography, and identified by MALDI-TOF mass spectrometer. Biological assays demonstrated that QUB-2607 exhibited different degrees of antibacterial activity against Staphylococcus aureus (S. aureus), Escherichia coli (E. coli), and Candida albicans (C. albicans), with minimum inhibitory concentrations (MIC) of 128 μM, 4 μM, and 128 μM, respectively. In MTT anticancer cell proliferation assays, QUB-2607 showed no significant anticancer activity against the NCI-H838 cell line across a concentration range from 0.001 μM to 100 μM. Furthermore, haemolysis tests on horse red blood cells showed no haemolytic activity even at the highest tested concentration of 256 µM. Based on these findings, QUB-2607 holds promise as a potent antimicrobial agent, particularly against E. coli, and has potential applications in developing new treatments for bacterial infections with minimal cytotoxic effects on mammalian cells. However, further research is needed to explore its full therapeutic potential and spectrum of activity.

Thesis is embargoed until 31st December 2029.
</dc:description>
